Research Progress on Chitinase and Chitosanase with Transglycosylation Activity

  • 摘要: 高聚合度的几丁寡糖和壳寡糖因其优越的功能活性而备受关注,但传统的制备方法存在诸多限制,如过程难以控制、分离纯化难等。近年来的研究发现,部分几丁质酶和壳聚糖酶不仅具备水解活性,还具有转糖基(transglycosylation,TG)活性,TG反应生产高聚合度寡糖时表现出底物易获得、生产成本低等优势。本文综述了近年来具备TG活性的几丁质酶和壳聚糖酶的研究进展,分析了影响几丁质酶TG活性的关键氨基酸位点,并探讨了通过对这些位点进行分子改造后对TG活性的影响。旨在为深入研究具TG活性的几丁质酶和壳聚糖酶的作用机制及其应用开发提供参考,以进一步推动高聚合度寡糖的绿色制备技术和产业化发展。

     

    Abstract: N−acetyl chitooligosaccharides and chitooligosaccharideswithhigh degree of polymerization are highly valued for their superior bioactivities. However, traditional preparation methods face various limitations, such as uncontrollable processes and challenges inseparation and purification. Recent studies have discovered that certain chitinases and chitosanases possess both hydrolysis and transglycosylation (TG) activities. The TG reaction offers advantagesin producingoligosaccharides with high-degree polymerization, including easy accessibility to substrates and low production costs. This paper reviews recent advances in chitinases and chitosanases with TG activity, analyzes the key amino acid residues affecting TG activity in chitinases, and discusses the impact of molecular modifications of these key residues on TG activity. The goal is to provide insights for further research on chitinases and chitosanases with TG activity and their subsequent development and utilization, thereby promoting the green production and industrialization of oligosaccharides with high-degree polymerization.
