不同淡水鱼来源弹性蛋白肽的制备及性质分析

Preparation and Property Analyses of Elastin Peptides from Different Freshwater Fish Sources

  • 摘要: 为充分利用淡水鱼资源和开发新来源的弹性蛋白肽,本研究对三种常见大宗淡水鱼的整颗心脏及动脉球基本信息进行测定,包括其质量、冻干动脉球中总氮含量及锁链素(desmosine,Des)和异锁链素(isodesmosine,Ide)含量等,并以不同淡水鱼来源的动脉球为原料酶解制备弹性蛋白肽,对弹性蛋白肽中Des和Ide含量、白度、氨基酸组成、肽分子量分布、抗氧化能力等理化特性进行分析。结果表明:鳙鱼动脉球的质量最大,平均为1.26 g/个,显著大于草鱼和鲢鱼动脉球的质量(P<0.05),鲢鱼冻干动脉球的总氮含量最高为16.07%,草鱼、鲢鱼和鳙鱼冻干动脉球中Des和Ide总含量在0.36%~0.38%之间,三种鱼之间没有显著差别(P>0.05)。利用动脉球制备的三种弹性蛋白肽中,草鱼弹性蛋白肽的得率最高,为6.01%,鲢鱼弹性蛋白肽的得率最低,为4.13%,鲢鱼弹性蛋白肽中的Des和Ide总含量最高,为0.33%,显著高于其他两种弹性蛋白肽中的含量(P<0.05)。三种弹性蛋白肽的氨基酸组成中,甘氨酸、丙氨酸、谷氨酸、脯氨酸和缬氨酸是含量较高的氨基酸,尤其是草鱼弹性蛋白肽中甘氨酸含量高达26%,组氨酸和蛋氨酸含量均较低,而羟脯氨酸含量没有显著差异(P>0.05),在2.36%~2.41%之间,鳙鱼弹性蛋白肽中必需氨基酸含量最高,为27.77%。此外,鳙鱼弹性蛋白肽中分子质量小于500 Da的肽段占比最高,达到75.74%,草鱼弹性蛋白肽的ABTS+自由基清除能力最强,在质量浓度为2.0 mg/mL时,对ABTS自由基的清除率高达93.54%。结果显示,淡水鱼动脉球是制备弹性蛋白肽的潜在优势原料,不同品种淡水鱼弹性蛋白肽的性质存在一定差异。本研究为淡水鱼资源的充分利用及开发高质量的弹性蛋白肽产品提供了理论依据。

     

    Abstract: In order to fully utilize freshwater fish resources and develop new sources of elastin peptides, the basic information of the whole heart and bulbus arteriosus of three common bulk freshwater fish was measured in this study, including their mass, total nitrogen content in freeze-dried bulbus arteriosus, and content of desmosine (Des) and isodesmosine (Ide). Elastin peptides were prepared by enzymatic hydrolysis of bulbus arteriosus from different freshwater fish sources. Analyses of physicochemical properties of elastin peptides were conducted, such as the content of Des and Ide, whiteness, amino acid composition, peptide molecular weight distribution, and antioxidant capacity. The results showed that the average weight of the bulbus arteriosus of bighead carp was the highest, at 1.26 g/bulb, which was significantly larger than that of grass carp and silver carp arterial bulbs (P<0.05). The total nitrogen content of the freeze-dried bulbus arteriosus of silver carp was the highest, at 16.07%, while the total content of Des and Ide of the freeze-dried bulbus arteriosus ranged from 0.36% to 0.38%, with no significant difference among the three fish species (P>0.05). Among the three prepared elastin peptides, the yield of grass carp elastin peptides was the highest at 6.01%, while the yield of that for silver carp was the lowest at 4.13%. The total content of Des and Ide of the silver carp elastin peptide was the highest at 0.33%, which was significantly higher than that of the other two elastin peptides (P<0.05). Among the amino acid compositions of the three elastin peptides, glycine (Gly), alanine (Ala), glutamic acid (Glu), proline (Pro), and valine (Val) were the amino acids with higher content. Especially for grass carp elastin peptides, Gly content was as high as 26%, while histidine (His) and methionine (Met) contents were relatively low. However, there was no significant difference in hydroxyproline (Hyp) content (P>0.05), ranging from 2.36% to 2.41%. The essential amino acid contents of bighead carp elastin peptide were the highest, up to 27.77%. In addition, the proportion of peptides with molecular weight less than 500 Da in the elastin peptide of bighead carp was the highest, reaching 75.74%. Grass carp elastin peptides had the strongest ability to scavenge ABTS+ free radicals, with a clearance rate of 93.54% at a mass concentration of 2.0 mg/mL. The results show that freshwater fish bulbus arteriosus are potential advantageous raw materials for preparing elastin peptides, and there are certain differences in the properties of elastin peptides derived from different varieties of freshwater fish. The research provides a theoretical basis for the full utilization of freshwater fish resources and the development of high-quality elastin peptide products.
