黑果腺肋花楸多酚的提取、鉴定及降糖降脂活性研究

Extraction, Identification and Hypoglycemic and Lipid-lowering Activity of Polyphenols from Aronia melanocarpa

  • 摘要: 探究超声波辅助低共熔溶剂(Deep eutectic solvents)提取黑果腺肋花楸浆果多酚(Aronia melanocarpa Polyphenols,AMP)的提取工艺、组成及降糖降脂活性。通过单因素实验和Box-Behnken响应面法分析得到最佳工艺,并采用超高效液相色谱-质谱/质谱(Ultra Performance Liquid Chromatograph Mass Spectrometer,UPLC-QTof-MS/MS)技术对经大孔树脂纯化后的AMP组分进行鉴定分析,最后通过体外模拟胃肠道的消化性,测定了其对α-葡萄糖苷酶及胰脂肪酶抑制能力。结果表明 DES-2(氯化胆碱-乳酸)摩尔比1:3、含水量80%、超声功率240 W、超声时间40 min为最佳提取条件,AMP得率最高为51.27 mg/g;DES提取的AMP纯化物中共有45种成分,包括15种花色苷、3种原花青素、18种类黄酮、9种酚酸,总含量为116.92±7.54 mg/g,其组成数量及含量均显著高于传统有机溶剂提取的纯化物。经口腔、胃、肠消化后AMP纯化物对α-葡萄糖苷酶抑制率达到30.4%±1.2%,是蓝莓多酚纯化物的3.34倍(P<0.05),胰脂肪酶抑制率达到92.7%±2.9%,是蓝莓多酚纯化物的1.04倍(P<0.05)。DES提取AMP相比传统有机溶剂提取效率高、产量高、无污染、组分多、活性强,具有一定的食品工业应用可行性。

     

    Abstract: The extraction process, composition and hypoglycemic and lipid-lowering activity of Aronia melanocarpa polyphenols (AMP) extracted by ultrasonic-assisted deep eutectic solvents were investigated. The optimal process was obtained by single factor experiment and Box-Behnken response surface method. The AMP components purified by macroporous resin were identified and analyzed by ultra performance liquid chromatograph mass spectrometer (UPLC-QTof-MS/MS). Finally, the inhibitory ability of α-glucosidase and pancreatic lipase was determined by simulating the digestibility of gastrointestinal tract in vitro. The results showed that the molar ratio of DES-2 (choline chloride-lactic acid) was 1:3, the water content was 80%, the ultrasonic power was 240 W, and the ultrasonic time was 40 min. The highest yield of AMP was 51.27 mg/g. There were 45 components in the purified AMP extracted by DES, including 15 anthocyanins, 3 proanthocyanidins, 18 flavonoids, and 9 phenolic acids. The total content was 116.92±7.54 mg/g, and its composition and content were significantly higher than those extracted by traditional organic solvents. After oral, gastric and intestinal digestion, the inhibition rate of AMP purified product on α-glucosidase reached 30.4%±1.2%, which was 3.34 times that of blueberry polyphenol purified product (P<0.05), and the inhibition rate of pancreatic lipase reached 92.7%±2.9%, which was 1.04 times that of blueberry polyphenol purified product (P<0.05). Compared with traditional organic solvents, DES extraction of AMP has high extraction efficiency, high yield, no pollution, many components and strong activity, and has certain feasibility of food industry application.
