酸/碱提取菠萝蜜果丝多糖及其结构、生物活性的影响

Effect of Acid/Alkali Extraction of Polysaccharides from Jackfruit Filum and Its Structure and Biological Activity

  • 摘要: 本实验以菠萝蜜果丝为研究对象,分别采用酸提取法、碱提取法,制备菠萝蜜果丝多糖。测定多糖得率,利用高效液相色谱、红外光谱、离子色谱、核磁共振、差示扫描量热等技术对多糖结构进行分析和比较。利用体外抗氧化、降血糖活性实验分析比较多糖的生物活性。结果表明,酸提多糖(AcJFP)得率小于碱提多糖(AlJFP)。2种多糖均由葡萄糖、半乳糖醛酸等单糖组成;AcJFP、AlJFP的重均分子量分别是1.01 kDa和12.08 kDa;均含有αβ构型的糖苷,存在较长的侧链和较多的分支,不存在三螺旋结构;X射线衍射谱显示2种多糖的晶体聚集结构存在差异;热力学显示均具有良好的热稳定性。体外生物活性表明,AcJFP具有更强的DPPH、羟基、ABTS+自由基清除能力、总抗氧化能力、α-淀粉酶活性抑制能力,AlJFP具有更强的亚铁离子螯合能力。因此,酸提与碱提对菠萝蜜果丝多糖的结构与生物活性存在影响。

     

    Abstract: This experiment focused on the preparation of polysaccharides from jackfruit filum using both acid and alkaline extraction methods. After determining the polysaccharide yield, the structures of the polysaccharides were analyzed and compared using various techniques, including high-performance liquid chromatography, Fourier-transform infrared spectroscopy, ion chromatography, nuclear magnetic resonance, and differential scanning calorimetry. The biological properties of the polysaccharides were evaluated through in vitro assessments of their antioxidant and hypoglycemic activities. The results indicated that the yield of acid-extracted polysaccharide (AcJFP) was lower than that of alkali-extracted polysaccharide (AlJFP). Both AcJFP and AlJFP fractions were composed of monosaccharides such as glucose and galacturonic acid. The weight-average molecular weights were 1.01 kDa for AcJFP and 12.08 kDa for AlJFP. Both fractions contained α and β glycosidic linkages, together with long side chains and numerous branches, and lacked a triple-helical structure. X-ray diffraction patterns revealed differences in the crystalline aggregation structures of the two polysaccharides, while thermogravimetric analysis demonstrated good thermal stability. In vitro biological activity assessments showed that AcJFP exhibited stronger DPPH, hydroxyl, ABTS+ radical-scavenging abilities, total antioxidant capacity, and inhibition of α-amylase activity, while AlJFP showed superior ferrous ion-chelating ability. Therefore, the method of extraction impacts both the structure and biological activities of polysaccharides from jackfruit filum.
