营养素影响蛋白质-EGCG非共价物体外抗衰老活性的研究

Effects of Nutrients on Anti-aging Activities in Vitro of Protein-EGCG Non-covalent Complex

  • 摘要: 为研究营养素添加对蛋白质-表没食子儿茶素没食子酸酯(EGCG)非共价物体外抗衰老活性的影响,本文构建了四类蛋白-EGCG非共价物(大豆分离蛋白-EGCG非共价物、酪蛋白-EGCG非共价物、鸡蛋清白蛋白-EGCG非共价物和β乳球蛋白-EGCG非共价物),以酪氨酸酶、胶原蛋白酶、弹性蛋白酶、透明质酸酶抑制率为体外抗衰老指标,比较四种蛋白-EGCG非共价物的体外抗衰老活性,筛选出蛋白质-EGCG非共价物载体,单一或复合添加大豆油、蔗糖和钙等3种营养素,探讨营养素添加对蛋白质-EGCG非共价物体外抗衰老活性的影响。结果显示,大豆分离蛋白-EGCG非共价物的酪氨酸酶、弹性蛋白酶和胶原蛋白酶抑制率整体高于其它非共价物,可作为载体进行营养素添加;营养素添加后,大豆分离蛋白-EGCG非共价物体系的酪氨酸酶、胶原蛋白酶和弹性蛋白酶抑制率显著升高(P<0.05),其中,弹性蛋白酶抑制率增幅最高,胶原蛋白酶次之,酪氨酸酶最低,且这种作用效果因营养素添加种类和组成而异。总的来讲,蛋白质与EGCG非共价结合可一定程度上改善EGCG的体外抗衰老活性,蔗糖、钙和大豆油添加后有助于大豆分离蛋白-EGCG体外抗衰老活性的提高。

     

    Abstract: In this paper, four protein-EGCG non-covalent complexes were prepared by EGCG and protein (soybean protein isolate, casein, egg albumin and β lactoglobulin) to investigate effects of nutrient supplementation on the anti-aging activity of protein-epigallocatechin gallate (EGCG) non-covalent complex. With the inhibiting rates of tyrosinase, collagens, elastase and hyaluronidase as the anti-aging indicators in vitro, the anti-aging properties of four protein-EGCG non-covalent complexes were compared to select the proper carrier for exploring effects of nutrient addition (single or compound addition of soybean oil, sucrose and calcium) on the anti-aging activities of protein-EGCG non-covalent complex. The results showed that soybean protein isolate-EGCG non-covalent complex had higher inhibitory rates of tyrosinase, elastase and collagenase than others, which would be significantly increased after nutrient supplementation (P<0.05). Among them, the increase of elastase inhibitory rate was highest, followed by collagenase inhibitory rate, and tyrosinase inhibitory rate was the lowest. Effects of nutrients on anti-aging activities in vitro of protein-EGCG non-covalent complex varied with the type and composition of nutrients. In general, the anti-aging activity of EGCG could be improved after its non-covalent binding to protein, the addition of sucrose, calcium and soybean oil into protein-EGCG non-covalent complex was prone to have superior anti-aging activity in vitro.
