刺梨天麻果脯发酵液对失眠小鼠的改善作用

Improvement of the Fermentation Liquid of Rose roxburghii and Gastrodia elata Fruit Preserves on Insomnia in Mice

  • 摘要: 本实验以刺梨天麻果脯发酵液(Rose roxburghii and Gastrodia elata fermentation liquid,RGFL)为主要材料,探究RGFL对DL-4氯苯丙氨酸(DL-4-chlorophenylalanine,PCPA)诱导睡眠剥夺小鼠的改善作用。连续3 d采用腹腔注射450 mg/kg的PCPA混悬液诱导小鼠失眠模型,进行戊巴比妥纳诱导小鼠睡眠实验,旷场试验,测定血清中超氧化物歧化酶(Superoxide Dismutase,SOD)、丙二醛(Malondialdehyde,MDA)、白细胞介素-6(Interleukin-1,IL-6)、白细胞介素-1β(Interleukin-1β,IL-1β)、肿瘤坏死因子α(Tumor necrosis factor-α,TNF-α)的水平以及测定脑组织中谷氨酸(Glutamic acid,Glu)、γ-氨基丁酸(Gamma-aminobutyric acid,GABA)、5-羟色氨酸(5hydroxytryptamine,5-HT)、多巴胺能(Dopaminergic,DA)含量变化来探究RGFL对失眠小鼠的改善效果,最后通过全脑组织病理切片评价RGFL对脑损伤的逆转作用。结果显示,利用阈上剂量(45 mg/kg)的戊巴比妥钠诱导小鼠睡眠后,Model组小鼠的入睡率仅为70%,睡潜伏期(6.33±1.0 min)延长,而睡眠持续时间(23.20±17.13 min)缩短,RGFL干预可以显著(P<0.05)提高PCPA失眠小鼠的入睡率(100%),缩短其入睡潜伏期(3.29±1.98 min)并延长睡眠持续时间(82.0±19.64 min);矿场试验发现,RGFL还可以减少PCPA失眠小鼠兴奋、狂躁样行为;不仅如此,RGFL可以显著(P<0.05)抑制失眠小鼠血清中炎症因子和氧化损伤产物水平,使IL-6、IL-1β、TNF-α、MDA的水平显著(P<0.01)降低34.87%、24.13%、25.72%、45.74%,SOD酶活显著(P<0.01)升高50.18%;能够显著(P<0.01)下调脑组织中神经递质Glu(39.62%)的含量,上调GABA(66.84%、)、5-HT(63.37%)、DA含量(88.06%);病理切片表明RGFL可以减轻和改善海马区神经元细胞排列松散、水肿、细胞核凝固和空泡化现象,能在一定程度逆转脑损伤。所以,RGFL对PCPA诱导睡眠剥夺的小鼠具有一定的改善作用,且改善效果优于刺梨发酵液(Rose roxburghii fermentation liquid,RFL)、天麻发酵液(Gastrodia elata fermentation liquid,GFL)单独给药组,显示出协同增效的作用。

     

    Abstract: In this experiment, Rose roxburghii and Gastrodia elata fermentation liquid (RGFL) was used as the main material to investigate the effects of RGFL on DL-4-chlorophenylalanine (PCPA)-induced sleep deprivation in mice. A mouse model of insomnia was established by intraperitoneal injection of 450 mg/kg PCPA suspension for three consecutive days. Several assessments were conducted, including the pentobarbital sodium-induced sleep test, open field test, and the measurement of serum levels of superoxide dismutase (SOD), malondialdehyde (MDA), interleukin-6 (IL-6), interleukin-1β (IL-1β), and tumor necrosis factor-α (TNF-α). Additionally, changes in the brain levels of glutamic acid (Glu), gamma-aminobutyric acid (GABA), 5-hydroxytryptamine (5-HT), and dopamine (DA) were analyzed to evaluate the effects of RGFL. Finally, histopathological examination of whole-brain tissue was performed to assess the reversal of brain damage by RGFL. The results showed that, after inducing sleep in mice using a suprathreshold dose (45 mg/kg) of sodium pentobarbital, the model group mice had a sleep onset rate of only 70%, a prolonged sleep latency (6.33±1.0 min) and a shortened sleep duration (23.20±17.13 min), and that the RGFL intervention significantly (P<0.05) increased the sleep onset rate (100%), shortened the sleep latency (3.29±1.98 min) and prolonged the sleep duration (82.0±19.64 min) of the insomnia-prone mice in the PCPA group. The open field test revealed that RGFL reduced hyperactivity and manic-like behaviors in insomnia mice. Furthermore, RGFL significantly (P<0.01) decreased the levels of IL-6, IL-1β, TNF-α, and MDA by 34.87%, 24.13%, 25.72%, and 45.74%, and significantly (P<0.01) increased SOD enzyme activity by 50.18%, and it was able to significantly (P<0.01) down-regulate the content of neurotransmitter Glu (39.62%) and up-regulate the content of GABA (66.84%,), 5-HT (63.37%), DA content (88.06%). Histopathological analysis demonstrated that RGFL alleviated and improved pathological features in the hippocampal region, such as neuronal disorganization, edema, nuclear pyknosis, and vacuolization, partially reversing brain damage. In conclusion, RGFL effectively improved sleep deprivation induced by PCPA in mice, showing superior efficacy compared to the single administration of Rose roxburghii fermentation liquid(RFL) or Gastrodia elata fermentation liquid (GFL) alone. This indicates a synergistic effect.
