羟基磷灰石稳定的岩藻黄质Pickering乳液制备及其对金枪鱼的保鲜作用

Preparation of Fucoxanthin-loaded Pickering Emulsion Stabilized by Hydroxyapatite and the Preservation Effect of Tuna

  • 摘要: 本研究以贻贝壳为原料制备的羟基磷灰石(hydroxyapatite,HAp)为稳定剂,制备HAp-Pickering乳液(HAp-PE)。探究不同油水比和HAp质量分数对HAp-PE稳定性、流变性能的影响,根据实验结果筛选出的最优条件制备HAp-PE并包埋岩藻黄质(fucoxanthin,Fx),得到HAp-PE-Fx。随后,研究了不同Fx添加量对HAp-PE-Fx抗氧化和抗菌效果的影响,选择合适的Fx添加量制备HAp-PE-Fx并用于金枪鱼鱼片的保鲜。结果表明:当油水比为60:40、HAp质量分数为6%时,HAp-PE的粒径最小(49.71 µm),乳化指数最大,表观黏度最高,说明其稳定性最佳;Fx浓度为0.75 mg/mL时,HAp-PE-Fx具有良好的抗氧化和抗菌(大肠杆菌和金黄色葡萄球菌)活性。此外,HAp-PE-Fx可有效保持金枪鱼鱼片肉色,降低贮藏期间金枪鱼鱼片的pH、总挥发性盐基氮含量和硫代巴比妥酸值,抑制微生物的繁殖。说明在4 ℃冷藏条件下,HAp-PE-Fx能有效延缓金枪鱼鱼片的腐败变质,保鲜效果明显。

     

    Abstract: In this study, hydroxyapatite (HAp) derived from mussel shells was used as stabilizer to prepare HAp-Pickering emulsion (HAp-PE). The effects of different oil-to-water ratios and HAp mass fractions on the stability and rheological properties of HAp-PE were analyzed. Based on the experimental results, the optimal conditions were established for the preparation of HAp-PE loaded with fucoxanthin (Fx) to form HAp-PE-Fx. Subsequently, the influence of varying Fx concentrations on the antioxidant and antibacterial properties of HAp-PE-Fx was evaluated. The optimal Fx concentration was then used to prepare HAp-PE-Fx for the preservation of tuna fillets. Results indicated that an oil-to-water ratio of 60:40 and a HAp mass fraction of 6% yielded HAp-PE with the smallest droplet size (49.71 µm), the highest emulsification index, and the greatest apparent viscosity, signifying optimal stability. At a Fx concentration of 0.75 mg/mL, HAp-PE-Fx demonstrated excellent antioxidant activity and antibacterial activity against Escherichia coli and Staphylococcus aureus. Furthermore, HAp-PE-Fx effectively preserved meat color as well as reduced pH, total volatile basic nitrogen content and thiobarbituric acid reactive substance value, inhibited microbial growth in tuna fillets during storage. These findings suggest that HAp-PE-Fx effectively retards tuna spoilage at 4 ℃, presenting an advantageous preservation effect.
