基于主成分分析的山药发酵菌株筛选

Study on Strain Screening for Yam Fermentation Based on Principal Component Analysis

  • 摘要: 本研究旨在筛选出能够高品质发酵山药的益生菌。以8株益生菌为供试菌种,分析发酵前后山药的酸度、活菌数、总黄酮、总多糖、尿囊素含量及DPPH自由基清除率的变化,并结合主成分分析法进行综合评价。结果表明,8株供试菌株均能显著增加发酵后山药中的乳酸、乙酸、总多糖和薯蓣皂苷元含量(P<0.05),其中植物乳植杆菌P9发酵后总黄酮含量最高,为0.208 g/L,比未发酵样品提高了39%;薯蓣皂苷元含量也以植物乳植杆菌P9发酵组最高(0.290 g/L);此外,嗜酸乳杆菌JZ1004和植物乳植杆菌P9能显著增强DPPH自由基清除能力(P<0.05)。主成分分析显示,植物乳植杆菌P9与干酪乳酪杆菌JZ1089的综合表现最优,在总黄酮、总多糖、尿囊素含量及抗氧化活性等方面均具有较高评分。本研究基于发酵性能、活性成分含量及抗氧化能力进行系统筛选,最终得到植物乳植杆菌P9和干酪乳酪杆菌JZ1089 2株适用于山药发酵的优良菌株,可为开发山药益生菌发酵饮品提供理论与菌种支撑。

     

    Abstract: This study aimed to screen probiotic strains suitable for high-quality fermentation of yam. Using 8 probiotic strains as test strains, the changes in acidity, viable bacteria count, total flavonoids, total polysaccharides, vitellin content, and DPPH free radical scavenging rate of mountain yam before and after fermentation were analyzed. Principal component analysis was employed for comprehensive evaluation. The results showed that all eight strains significantly increased the contents of lactic acid, acetic acid, total polysaccharides, and diosgenin in fermented yam (P<0.05). Among them, Lactiplantibacillus plantarum P9 yielded the highest total flavonoid content (0.208 g/L), which was 39% higher than that of the unfermented sample, and also the highest diosgenin content (0.290 g/L). In addition, Lactobacillus acidophilus JZ1004 and L. plantarum P9 significantly enhanced the DPPH radical scavenging ability (P<0.05). Principal component analysis indicated that L. plantarum P9 and Lacticaseibacillus casei JZ1089 performed best overall, with high scores in terms of total flavonoids, total polysaccharides, allantoin content, and antioxidant activity. Based on systematic screening considering fermentation performance, active component content, and antioxidant capacity, L. plantarum P9 and L. casei JZ1089 were identified as two excellent strains suitable for yam fermentation, providing theoretical and strain support for the development of yam-based probiotic fermented beverages.
