沉香叶成分分析及助睡眠作用

Analysis of Components of Agarwood Leaves and Study on its Sleep-inducing Effect

  • 摘要: 目的:本研究旨在探究沉香叶挥发性成分、促睡眠作用及可能作用机制。方法:利用水蒸气蒸馏法提取沉香叶水提物;采用气相色谱-质谱联用仪,检测沉香叶水提物中挥发性成分;选用经典对氯苯丙氨酸(p-Chlorophenylalanine,PCPA)诱导小鼠失眠模型,检测小鼠的睡眠情况、自主活动,评价沉香叶水提物的促睡眠作用;采用ELISA方法,检测血清中神经递质谷氨酸(Glutamic acid,Glu)、伽马-氨基丁酸(Gamma-aminobutyric acid,GABA)及5-羟色胺(5-hydroxytryptamine,5-HT)水平,探究沉香叶促睡眠的可能作用机制。结果:经分析鉴定,沉香叶含有挥发性成分65个,其主要成分(占比>1%)有21个(77.72%),为香叶基丙酮、顺-1-甲基双环癸烷-2,10-二酮、十六酸、6,10,14-三甲基-2-十五烷酮、顺式-3-己烯醇苯甲酸酯、反式-橙花叔醇、正辛醇、β-紫罗酮等化合物。药效研究显示,沉香叶水提物可显著缩短失眠小鼠入睡潜伏期(P<0.01),延长小鼠睡眠时间(P<0.01或P<0.001),有良好的促睡眠作用;沉香叶水提物明显降低失眠小鼠的运动总路程(P<0.01)和平均速度(P<0.01或P<0.001),延长静止时间(P<0.01),表现良好的抑制小鼠自主活动作用;沉香叶水提物还可显著降低失眠小鼠Glu水平,升高GABA和5-HT水平(P<0.05或P<0.01)。结论:研究表明,沉香叶水提物可助睡眠,作用机制与调节神经递质分泌和调控兴奋-抑制神经递质(Glu-GABA)分泌平衡有关。

     

    Abstract: Objective: This study aimed to investigate the sleep-promoting effect and possible mechanism of volatile components from agarwood leaves. Methods: The aqueous extract of agarwood leaves was obtained by steam distillation; the volatile components were detected by gas chromatography-mass spectrometry(GC-MS) in the aqueous extract of agarwood leaves; insomnia model induced by p-Chlorophenylalanine (PCPA) was used to evaluate the sleep-promoting effect in mice of the aqueous extract of agarwood leaves; the levels of neurotransmitters glutamic acid (Glu), gamma-aminobutyric acid (GABA), and 5-hydroxytryptamine (5-HT) were detected by ELISA to explore the possible mechanism of sleep-promoting effect of agarwood leaf in serum. Results: Sixty-five volatile compounds were identified from the agarwood leaves, among which the main components (accounting for >1%) were twenty-one compounds (77.72%), including cinnamyl acetone, cis-1-methyldicyclopentane-2,10-dione, hexadecanoic acid, 6,10,14-trimethyl-2-pentadecanone, cis-3-hexenol benzoate, trans-geraniol, n-octanol, β-ionone, etc. The pharmacological study showed that the aqueous extract of agarwood leaves could significantly shorten the sleep latency of insomnia model mice (P<0.01), prolong the sleep time (P<0.01 or P<0.001), and had a good sleep-promoting effect; the aqueous extract of agarwood leaves significantly reduced the total distance and average speed in insomnia model mice (P<0.01) and prolonged the rest time (P<0.01), showing good inhibitory effect on the spontaneous activity; the aqueous extract of agarwood leaves could also significantly reduce the Glu level and increase the GABA and 5-HT levels (P<0.05 or P<0.01). Conclusion: The study shows that the aqueous extract of agarwood leaves have goood promote sleep effect, and its mechanism is related to regulate the secretion of neurotransmitters and control the balance of excitatory-inhibitory neurotransmitters (Glu-GABA).
