火麻仁抗氧化肽的制备、分离纯化及体外消化吸收特性分析

Preparation, Separation, Purification, and in Vitro Analysis of the Digestion and Absorption Properties of Antioxidant Peptides from Hemp Seeds

  • 摘要: 本研究旨在筛选鉴定火麻仁高抗氧化性肽序列,并探究其胃肠道稳定性和小肠吸收透过性。以火麻仁蛋白为原料制备火麻仁肽(Hemp seed peptides,HSPs),并通过超滤、Sephadex G-15、液相色谱串联质谱技术(Liquid chromatography tandem-mass spectrometry,LC-MS/MS)筛选出高抗氧化性的肽序列,并进行了体外模拟胃肠道消化实验和Caco-2细胞单层模型转运实验。结果表明,经过超滤、Sephadex G-15分离后,F1组分的抗氧化性最强;LC-MS/MS结合虚拟筛选得到3条HSPs序列;体外胃肠道模拟消化实验和Caco-2细胞单层模型转运实验结果发现,HSPs序列Phe-Tyr(FY)的胃肠道稳定性良好,表观渗透系数(Apparent permeability coefficients,Papp)为(6.017±0.4998)×10−7 cm/s,属于中等吸收化合物。本研究为火麻仁蛋白的高值化利用提供新思路,为火麻仁特定肽序列经口摄入并发挥体内功效提供了科学依据。

     

    Abstract: This study aimed to screen and identify peptide sequences with potent antioxidant properties from hemp seeds and to evaluate their gastrointestinal stability and intestinal absorption permeability. Hemp seed protein was used as the raw material to prepare hemp seed peptides (HSPs). Peptides exhibiting high antioxidant activity were screened and isolated using a combination of ultrafiltration, Sephadex G-15 gel filtration, and liquid chromatography-tandem mass spectrometry (LC-MS/MS). Furthermore, in vitro simulated gastrointestinal digestion and Caco-2 cell monolayer transport assays were conducted. The results demonstrated that the F1 fraction exhibited the strongest antioxidant activity following ultrafiltration and Sephadex G-15 separation. Three HSPs sequences were identified through LC-MS/MS coupled with virtual screening. Among them, the in vitro simulated gastrointestinal digestion and Caco-2 cell transport assays demonstrated that the peptide Phe-Tyr (FY) displayed good gastrointestinal stability, with an apparent permeability coefficient (Papp) of (6.017±0.4998)×10−7 cm/s, classifying it as a moderately absorbed substance. This study proposes a novel strategy for the high-value utilization of hemp seed protein and offers scientific evidence supporting the potential in vivo efficacy of specific hemp seed peptides upon oral administration.
